Lo and Ber runes are easiest to build toward when every run serves a purpose. Start with a character that can kill priority targets quickly, then use D4 items to fill any serious gear gaps before spending valuable materials. Slow boss clears usually cost more than they return, so do not force content that your build cannot handle comfortably.
Choose the Right Boss Runs
Baal in the hell version of Worldstone Keep is a strong target on Torment XII and above. His encounters can provide high-tier runes and an Annihilus charm. Belial, found in the Sanctum of Lies, adds another useful farming option, especially when you also need Mythic gear, Mythic Seals, and Sparks. Focus on single-target damage, resource control, and movement. A clean two-minute run is worth far more than a messy attempt that takes twice as long.
Make Each Undercity Run Count
The Kurast Undercity becomes much more useful with the right War Plans upgrades. Put points into Gutter Filth and Bauble Day before entering. Then follow a simple routine.
Activate every beacon you pass.
Kill each Portal Prankster that appears.
Defeat the district boss and visit the Cabochon Merchant.
Bauble Day allows the merchant to appear after the boss when a Portal Prankster was defeated. The vendor can offer runes, gems, and Prime Evil Splinters for gold, so check the inventory every time.
Craft Instead of Chasing Luck
A direct Lo or Ber purchase is possible, but it is not a sensible plan by itself. Merchant refreshes cost 666 Forgotten Souls, and repeated attempts can empty your stock quickly. Buy useful intermediate runes whenever they appear, then transmute them in the Horadric Cube. Two Vex runes and a Flawless Emerald make Ohm. Two Ohm runes and a Flawless Diamond make Lo. Two Lo runes and a Grand Topaz make Ber. The process takes patience, but each purchase moves you closer to the result.

Improve the Material Supply
Use a Tribute of Harmony before opening the Undercity portal and select Bargain for Invocation Runes. This changes the reward pool and gives you more opportunities to collect Vex and Ohm runes. Seasonal dungeons opened with Chronicle of Nightmare Sigils are worth running as well, since they can provide extra runes and the gems needed for transmutations. Mix these activities around your playtime, and your stockpile will grow without depending on one lucky merchant visit; that steady approach also makes D4 items cheap easier to fit into your wider upgrade plan.
